the one you would put using the DVD connector will act as a slave, and the regular one becomes the master. The master is used to boot.

On the Hard Disk itself, there is usually a diagram depicting the jumper settings for Master, Slave, Cable select modes...

Just put the 2nd disk's Jumper into Slave mode, and then connect. It will surely act as a slave then..
 
Do you mean removing my DVD drive and attaching 2nd HDD to it ? if yes then how will Windows boot means there are 2 HDD's now and which one should I choose for booting ?

Yeah as Abhas said Master will boot the system. You can also check BIOS setting to see boot sequence.
 
Just for the record, there are USB-to-USB solutions available that allow you to transfer data between computers. They won't be as fast as setting the hard drive up as a slave, though, and you will also have to go out and actually purchase the cable.
